Research Abstract	本研究は、日本人における気管支喘息の感受性遺伝子が存在する染色体領域を同定することを目的とした。日本人の小児気管支喘息患者を発端者とする45家系、190人、63同胞対を対象とした。気管支喘息患者の発端者(45人)と同胞(53人)は全員、医師によって気管支喘息と診断され、かつ、ダニ特異的IgE抗体陽性の者に限った。ダニ特異的IgE値がRASTクラス2以上の人を、ダニ特異的IgE抗体陽性者と判定した。多型マーカーはHuman Screening Set(Research Genetics)を用いた。ABI PRISM 377 Autosequencerで泳動し、GeneScanプログラムおよびGenotyperプログラム(Perkin-Elmer)を用いて遺伝子型を決定した。多点連鎖解析はGENEHUNTERを二点連鎖解析はSPLINKを使用した。その他にTransmission disequilibrium tests(TDTs)をSIB-PAIRプログラムを用いて行った。その結果、20、21、X染色体を除いてすべての染色体で解析を終了し、7カ所の連鎖の可能性のある領域が検出された。5番、12番、13番染色体の連鎖領域はCaucasianでも連鎖を示唆する報告がでており、多くの民族でアトピー喘息に関わる遺伝子が存在している可能性は高いと推測される。5番染色体の連鎖領域は多くの候補遺伝子がクラスターが、12番染色体の連鎖領域にはインターフェロンγ遺伝子やStem Cell Factorなどの気管支喘息の候補遺伝子が、13番染色体の連鎖領域にはSTAT5a遺伝子が存在しており、今後、さらに密にマーカーを用いて連鎖、TDTより、連鎖の可能性のある領域を狭め、遺伝子の変異検索を行って、関わっている遺伝子の同定をする必要がある。精神分裂病家系は、一次スクリーニングが可能な100家系の収集を終えた。
