| Research Abstract |
コムギ葉を材料に葉緑体を単離・精製し,その破壊液を用いてRubiscoの分解について調べた。その結果,Rubiscoの大サブユニットが光照射下において37kDa,16kDaのペプチドへと断片化されることを見い出した。同様な図りに依存した分解はホウレンソウ,エンドウ葉から精製した葉緑体を用いた実験においても確認された。さらに単離葉緑体を用いて活性酸素消去系酵素の阻害剤のKCN,NaN_3存在下,光照射したところ同様なRubiscoの分解が見い出された。これらの活性酵素の関与を示唆するデーターを裏ずけるためRbiscoを精製し,注性酸素種の一つであるヒドロキシルラジカルにさらしたところ,葉緑体およびその磨砕液での分解と全て同様に分解された。また切断部位をN末端,C末端分析に供したところ,破砕液で見い出された断片化の部位を精製Rubiscoを活性酸素にさらした場合の部位は完全に一致した。またこの分解がカルボキシレーション反応の中間体アナログCABPにより阻害されることを見い出した
