Research Abstract

これまで我々はNOを介するグルタミン酸神経毒性がドーパミン(DA)ニューロンに対するMPP^+の選択的毒性発現に重要な役割を果たすこと、さらに、ドーパミンD2受容体アゴニストおよび培養線条体細胞条件培地(CM)エーテル抽出物がグルタミン酸神経毒性に対して保護作用を発現すること、さらにCM中の神経保護活性物質の一部はCMに添加されている牛胎仔血清(FCS)に由来することを報告してきた。そこで、本研究ではグルタミン酸-NO系により誘発されるDAニューロン死を抑制する内在性物質としてFCSおよびCM由来蛋白性神経保護活性物質に注目し、以下の研究を行った。1)FCS由来神経保護活性物質の精製と化学構造の解析:FCSのエーテル抽出物をアセトニトリル/水相糸、および、メタノール/水相系の2段階の逆相クロマトグラフィーにて分類することにより、NO神経毒性とグルタミン酸神経毒性の両者を強く抑制する分画を得た。その分画についてマススペクトリー解析を行ったところ、ほぼ単一物質にまで精製されており、分子量に255に大きなフラグメントピークを持つ分子量365ないし383の物質であることが確認された。MS/MS解析の結果、本化合物はステロイド骨格を持つこと、既存の生理活性ステロイドとは異なることなどが明らがになった。3位のケトンが水酸化されていることから水酸化ステロイドの一種であると推定している。2)エステラジオールの神経保護作用:ラット胎仔由来培養中脳細胞を用いて、抗酸化作用を持つステロイドの一つとしてエステラジオール(17β-esteradiol)のグルタミン酸神経毒性に対する作用を検討した。17β-esteradiolはDAニューロンおよび非DAニューロンにおけるグルタミン酸神経毒性をともに抑制したが、その神経保護作用はエストロゲン受容体拮抗薬および蛋白合成阻害薬の影響を受けなかった。さらに、不活性光学異性体の17α-esteradiolもグルタミン酸神経毒性を抑制した。一方、コルチコステロンおよびテストステロンはグルタミン酸神経毒性を抑制しなかった。17β-esteradiolは過酸化水素により誘発される神経毒性も抑制した。以上の結果はエステラジオールが活性酸素種による神経毒性を抑制することにより中脳DAニューロンに対する保護作用を発現することを示唆する。
