| Research Abstract |
1、 virescent遺伝子のクローニング
すでに表現型マーカーとして知られている11のvirescent遺伝子(Vl-Vl1)のうち、突然変異株の表現型解析が進んでいるVl〜V3の3つについて、その変異に対するRFLPマッピングを開始した。vl〜v3突然変異株はいずれもJaponicaバックグラウンドのため、代表的なIndica系統であるKasarath系統と交雑して第一代雑種Flを作り、さらにこれを自殖して約6000個体からなるF2世代集団を得た。得られたF2世代集団よりvirescent表現型を示すホモ個体を、v1、v2、v3それぞれから選抜した。現在、各個体からDNAを抽出し、プールドサンプリング法を併用してサザンブロット解析によるRFLPマッピングを行っている。
2、 イネにおける核コードの葉緑体局在RNAポリメラーゼ遺伝子の解析
Arabidopsis rpoZ遺伝子、及び他の植物種においてミトコンドリア局在性のT3/T7型RNAポリメラーゼをコードすると推定されている遺伝子の塩基配列情報を基に、これらの遺伝子間で相同性の高い領域からdegenerate primerを作製した。このprimerを用いてイネのゲノムDNAからPCR反応を行ったところ、いくつかのDNA断片が増幅された現在、それらのDNA断片の塩基配列を調べているところである。
