Research Abstract	私共はアドレノメデュリン(AM)の血管作用は単に血管拡張だけでなく、血管平滑筋細胞(VSMC)の増殖促進作用を持ち、AMが血管リモデリングに関与している可能性を示した。本年度はAMの作用機序の解析ならびに生体内でのAMの作用の有無の検討を行った。ラットVSMCではAMはCa^<2+>応答性に細胞質チロシンキナーゼであるPYK-2とc-Srcを活性化し、アダプター蛋白(Shc、Grb-2-Sos)をリクルートする結果、ras依存性にMAPキナーゼ(ERK1/2)を活性化することが明らかとなった。更にラット頚動脈のバルーン傷害モデルでは新生内膜のVSMCにAMが過剰発現していることを免疫組織学的に確認した。更にバルーン傷害前よりAM受容体拮抗薬のCGRP(8-37)を2週間連続投与した結果、内膜肥厚部位は対照に比較して有意に減少していた。以上の事実は生体内でもAMは内膜傷害時に血管局所で過剰に発現され、AMがVSMCの遊走・増殖に促進的に作用していることを実証した。現在AMレセプターの補助蛋白であるRAMPアイソフォームを特異的に阻害するアンチセンス導入により、AMによる血管肥厚の抑制効果が生ずるかを検討中である。
