| Research Abstract |
造血幹細胞には、その増殖と文化を可能にする微小環境(ニッシェ)が与えられている。本研究は、造血幹細胞の微小環境の分子基盤を、(1)造血の発生、(2)造血幹細胞の移動・増殖の3点から明かにしようとするものである。
(1) 哺乳類の個体発生では、胎児卵黄嚢における胚型赤血球の産生(一次造血)によって造血が開始される。その後、AGM(Aorta:大動脈、Conad生殖器、Mesonephros:中腎)領域において自己複製能を有する造血幹細胞が発生し、その造血幹細胞が卵黄嚢、あるいは胎児肝・骨髄へと移行し、二次造血が営まれるようになる。本年度、われわれは、造血幹細胞と血管内皮細胞の共通祖先細胞であるHemangioblastの同定を行い、マウス造血幹細胞が胎生8-9日の間に、背側大動脈などの血管内に発生することを明らかにした。
(2)AGMに発生した幹細胞は、受容体型チロシンキナーゼTIE2とその結合因子であるAngiopoietinのシグナルよってインテグリンが誘導され、それが幹細胞の増殖に関わることを明らかにした。
