| Research Abstract |
前年度の研究により、テロメレース活性は触媒サブユニットTERTの発現制御により主に規定されていることが明らかにされた。そこで、本年度においては、生体内でTERTの発現を示しおそらくテロメレース陽性と思われる細胞を可視化するために、mTERTのプロモーター約10kbをb-galactosidase-neoのコード領域につなげたレポーター遺伝子を作成し、その発現パターンを観察した。テロメレース陽性細胞は、増殖する未分化な細胞と考えられるので、このようにレポーター遺伝子で可視化した細胞は、組織幹細胞や前駆細胞を同定することに役立つと期待される。先ず、レポーター遺伝子をマウス胎児性幹細胞ES細胞に遺伝導入し、フィーダーを除きembryoid bodyを作らせて細胞を分化させた時のレポーターの発現パターンを解析した。すると、細胞分化に伴って内因性のmTERT発現が低下しテロメレース活性が失われると平行してb-galactosidase活性が低下し、レポーター遺伝子が内在性TERT遺伝子のレポーターとして期待通りに機能することを確認した。そこで、このレポーター遺伝子のトランスジェニックマウスを作成した。得られたマウスの組織を予備的に解析したところでは、期待通り幹細胞が存在すると思われる部位にb-galactosidase遺伝子の発現が認められた。
