Research Abstract	トランスジェニック(Tg)家畜の作出で重要な課題は、Tg動物の作出効率を上げることである。本研究はEGFPをマーカーに利用し、Tg家畜を効率に作出するための基礎研究として実施した。マーカー用EGFP遺伝子と目的の遺伝子とを連結した場合のTg胚の選抜効率、Tg動物の作出効率、目的遺伝子の発現等について検討した。全身性の発現があるCAGプロモーター/EGFP融合遺伝子の3'側に有用遺伝子としてWAPプロモーター/hGH融合遺伝子を連結したconstruct作成した。このCAG/EGFP-WAP/hGH連結遺伝子をマウスの受精卵前核に顕微注入し、生存胚を体外培養した。その後、胚盤胞時に蛍光顕微鏡で観察し、全体蛍光胚、部分蛍光胚、蛍光胚の胚の3種類に選別し移植した。比較用に選別をしない胚の移植も行った。生まれた個体マウスの尾部から抽出したDNAを用いてPCRによりTg個体を同定した。また、成体Tgマウスから採血し、血中のhGH量を調べた。Tg胚の選抜効率に影響なく、極めて高率にTg個体が得られた。また、全身性にEGFPの発現が確認されたTg個体の全てにおいて血中にhGHが検出された。さらに、泌乳中のTgマウスから乳中を採取し、現在hGH量の測定中である。
