| Research Abstract |
進行性多巣性白質脳症(progressive multifocal leukoencephalopathy:PML)はポリオーマウイルスに属する2本鎖環状DNAウイルスであるJCウイルス(JCV)が原因である。しかしながらJCVがヒト脳の脱髄疾患であるPMLを引き起こす詳細なメカニズムは不明であり、根治的な治療法は全くない。治療法確立の前段階として、我々はJCVの感染および増殖メカニズムを解明するために、
1)ウイルス受容体の同定、単離、
2)ウイルス転写活性化機構の解析を試みた。
ウイルス受容体を同定するために、JCウイルス外郭蛋白 VP1 を C_<OS>7 細胞で発現させることにより、recombinant偽ウイルス粒子作成法を確立した。偽ウイルス粒子は電子顕微鏡的にウイルス粒子構造が確認され、また、bona fide粒子同様に赤血球凝集活性を有していた。偽ウイルス粒子は内部にゲノムをもたないため外来遺伝子を挿入することが可能であり、我々の開発したjcウイルスベクターは受容体単離のみならず神経系組織への遺伝子治療を目的とするベクターとして使用可能である。
転写活性化機構の解析については、HTLV-I 感染者に発症したPMLの解析から、Tax蛋白に着目し、Taxがnauroglial cell においてJCウイルスの転写を活性化することを明らかにした。また、JCウイルス転写能の神経特異性についてはTax共沈する未知の蛋白を同定し現在論文投稿中である。
