| Research Abstract |
幼若ホルモン(JH)は昆虫の変態・休眠・相変異など特異な生理現象の調節に関わっている。しかしJH受容体の実体はじめそのシグナル伝達の詳細は不明である。この研究ではJH応答性転写調節因子の同定とクローニング更にはJH受容体の同定を行なって、JH-情報伝達系を解明することを目的とした。
Cyanoprotein遺伝子のJH応答配列(JHRE)とJH処理脂肪体cDNAライブラリーを用いて、以下のような実験を組み合わせることでJH応答性転写調節因子(JHRTF)をクローニングすることを試みた。
1)発現ベクター又はphageに導入したcDNAライブラリーとプローブとしてラベルしたJHREを用い、South-Western法・phage-display法によりスクリーニングする。
2)JHREとレポーター遺伝子とのDNAコンストラクト及びcDNA-Gal4 fusionライブラリーを酵母に導入し、one-hybrid法によりスクリーニングする。
3)cDNA-Gal4 fusionライブラリーと既知転写因子Gal4 fusion DNAコンストラクトとを用い、酵母two-hybrid法によりJH依存性の未知転写因子JHRTFをスクリーニングする。
これらのうち今年度に関してはone-hybrid法によるスクリーニングを行い候補となるクローンを複数個pick upし、現在それらについて解析中である。
