水分恒常性の維持機構の分子細胞生物学的研究(神経内分泌細胞間・内情報伝達の解析)

Research Abstract	視床下部室傍核および視索上核は水分の恒常性維持のための中枢として重要な部位である。これらの部位には抗利尿作用を有するバゾプレッシ産生細胞が存在する。この神経分泌細胞の活動は、液性・神経性に調節されている。そこで、種々の神経伝達物質や修飾物質の神経分泌細胞への直接作用および入力系への作用について検討した。さらに、神経分泌細胞自身はバゾプレッシンを産生するだけではなく、種々の神経ペプチドやNO合成酵素を含有しているので、その存在意義について検討した。得られた結果は以下の通りである。1、PACAPおよびプロスタグランデインE2はラット視索上核の神経分泌細胞の神経活動を非選択性陽イオンチャネルを開くことにより興奮させた。2、PACAPおよびATPははラット視索上核の神経分泌細胞の細胞内カルシウム濃度をそれぞれに特異的な受容体を介して増加させた。3、GABAはラット視索上核の神経分泌細胞のGABAB受容体を介して神経活動を抑制した。4、PACAPおよびアドレノメデュリンの覚醒ラット脳室内投与により室傍核および視索上核のバゾプレッシン産生細胞とオキシトシン産生細胞がそれぞれ特異的に活性化されることを明らかにした。5、ラット室傍核および視索上核の神経型NO合成酵素遺伝子の発現が正常浸透圧性容量減少刺激および糖尿病状態で増加していた。今後、視床下部における神経分泌細胞の特性を総合的に解析することにより水分の恒常性維持のための中枢としての役割を解明していきたい。
