| Research Abstract |
慢性関節リウマチ(以下RA)の関節軟骨破壊に軟骨細胞外マトリックスの分解は必須であり、一群のマトリックスメタロプロテアーゼ(MMP)の関与が近年明らかになってきた。本研究課題の目的は、RA関節軟骨破壊に及ぼすMMPの分子作用機構を明らかにすることである。本年度得られた研究成果は、以下の如くである。
(1).潜在型MMP-2活性化酵素のMT1-MMPとMT3-MMPは、MMPの組織インヒビター(TIMP-1,2,3)のうちTIMP-1では阻害されないことを明らかにするとともに、MT3-MMPも細胞外マトリックス成分を分解することを示した。また、MT1-MMPはある条件下で細胞膜よりsheddingされることを見出した。
(2).ゼラチンフイルムを用いたin situ zymography法を開発し、MT1-MMPにより活性化された潜在型MMP-2のゼラチン分解活性を癌組織内で直接検出することに成功した。
(3).RA滑膜組織においては潜在型MMP-2の活性化がみられ、MT1,2,3-MMPのうちMT1-MMPの作用により活性化されていることを明らかにした。また、MT1-MMPはRA滑膜表層細胞に局在し、in situ zymographyで同部にゼラチン分解活性が実証された。
(4).RAと変形性関節症の関節液中MMP(MMP-1,2,3,7,8,9,13)とTIMP(TIMP-1,2)産生量を測定するとともに、関節液中のメタロプロテアーゼ活性を測定することにより、RA関節液ではMMP量がTIMP量を上回っていることを示した。
(5).MMPに対する合成インヒビターを製薬企業と共同で開発し、MT1-MMPに対する有効なインヒビターを開発した。また、RA滑膜細胞をペントサンポリ硫酸で刺激すると、TIMP-3は転写後のレベルで特異的に産生亢進されることを明らかにした。
