| Research Abstract |
手術肺および死後4時間以内の剖検肺から得られた気道を用いて、気道上皮細胞および気管支腺(粘膜下腺細胞)を単離し、また各々の培養細胞を作製した。これらを用いて、次のような所見を得た。すなわち、1)Surfactant protein-A(SP-A)に対するモノクローナル抗体を用いての免疫組織的検索でSP-Aが気管支腺の漿液細胞に局在しており、気道上皮細胞にはみられなかった。さらに、気管支腺からのSP-Aの分泌をELISAにて確認した。SP-Aの遺伝子発現は気道上皮細胞にはみられず、気管支腺細胞にのみに認められた。さらに、対照の肺胞組織ではSP-A1とSP-A2の両者の遺伝子発現がみられたが、気管支腺細胞にはSP-A2のみの発現がみられた。気管支腺からのSP-Aは肺胞領域とは異なる機序で気道防御作用を示すことが考えられた。2)気道上皮細胞および気管支腺細胞にhumanβ-defensin 1(hBD-1)とhBD-2の遺伝子発現がみられたが、気管支腺が上皮に比べて5倍高い発現を示した。好中球エラスターゼおよびメサコリンにより気管支腺のhBD-2の遺伝子発現が増強したが、hBD-1の遺伝子発現は変化しなかった。すなわち、hBD-1はconstituetiveに、そしてhBD-2はinducibleに発現することが示された。3)気管支腺に於いてはムチン分泌と同じ刺激でSP-AおよびhBDの遺伝子発現の増強がみられたが、気道上皮ではこの傾向はみられなかった。
