Research Abstract	腸管膜血管モデルと大動脈血管モデルを用いて生体内に存在するアンギオテンシンIIを刺激剤として血管収縮時のフリーラジカルの関与について検討した。腸管膜血管モデル:実験1;腸間膜血管に血流が停止する濃度のアンギオテンシンIIを添加すれば血流は停止するが、この停止は1次的なもので、すぐ血流の再開通が起こる。この血流停止条件下でスーパーオキサイドジスムターゼを添加すると、停止に至る時間が延長し、再開通の時間が短縮した。実験2;腸管膜血管を摘出し、MCLAを添加した緩衝液と培養し、アンギオテンシンIIを添加した所、スーパーオキサイドの産生を示す発光が見られた。実験3;スーパーオキサイドジムスターゼはアンギオテンシンIIによる腸管膜血管の収縮を有意に減弱させた。以上よりアンギオテンシンIIによる腸管膜血管の収縮にはスーパーオキサイドが発生し、かつ血管収縮に関与している可能性があることが判明した。大動脈血管モデル:実験1;アンギオテンシンIIの添加によって起こる血圧上昇に対して、スーパーオキサイドジムスターゼは血圧上昇を減少させた。実験2;大動脈血管を摘出し、MCLAを添加した緩衝液と培養し、アンギオテンシンIIを添加した所、スーパーオキサイドの産生を示す発光が見られた。実験3;スーパーオキサイドジムスターゼはアンギオテンシンIIによる大動脈血管の収縮を減弱させた。以上より大動脈血管でもアンギオテンシンIIによる収縮時にスーパーオキサイドが発生し、血圧上昇に関与している可能性があることが判明した。今回の実験より血管からスーパーオキサイドが産生されるという極めて特異な知見を得た。スーパーオキサイドは細胞増殖作用があるという最近の報告もあり、今回の結果と合わせるとアンギオテンシンIIはスーパーオキサイドをメヂエイターとして血管平滑筋増殖を促進していることが示唆された。
