Research Abstract	本研究では、染色体異営の原因となるDNA二本鎖切断、エンドジョイニング、非相同組換えの起こるメカニズム及びDNA修復機構との関わりについて、大腸菌及び酵母のアッセイ系を用いて調べた。まず、大腸菌においては複製に関わるDnaBヘリケースと相同組換えに関わるExo VIII(RecE)エキソヌクレアーゼ及びRecTタンパク質が共に非相同組換えを促進することを示した。一方、二本鎖切断修復に関わるDNA結合タンパク質H-NSは非相同組換えを抑制することを示した。さらに、大腸菌を栄養飢餓の状態に置いたときにDNA二本鎖切断及び非相同組換えの亢進が起こること、一本鎖切断が塩基のアルキル化によって誘導されること、さらに、TagおよびAlkA DNAグリコシラーゼがこの組換えを抑制することを示した。従って、非相同組換えは種々の修復機構と密接に関わっていることを見出した。出芽酵母においては、DNAトポイソメラーゼII阻害剤が非相同組換えを誘導することを見出し、トポイソメラーゼの崩壊による二本鎖切断とエンドジョイニングを経由してDNAの再編成に至ると考えた。さらに、ウエルナー症候群原因遺伝子の産物WRNと会合するタンパク質WHIPを見出した。出芽酵母WHIPはWRNホモログSgs1と遺伝学的に相互作用があり、二重変異株はより早く老化することが分かった。分裂酵母のDNA複製に必須なHsk1キナーゼの温度感受性変異とrad21変異が合成致死性を示した。これらは、コヒーシンがDNA複製とも密接な関連を持つことを示した。
