| Research Abstract |
本研究は、粘膜免疫機構から、国家的課題である炎症性、アレルギー性消化管障害の病因病態の解明とその治療法の開発を目的に企画された。
1)消化管アレルギー実験動物の確立と、それを用いた粘膜機構に関する基礎研究からのアレルギー発症機序と粘膜傷害機序の解明。
BACB/Cマウス、STAT6^<-/->マウス、TCRγ^<-/->マウスなどを卵白アルブミン(OVA)で全身感作することにより、経口投与された食物抗原(OVA)に対して抗原特異的に大腸に限局したアレルギー反応とそれに引き続く下痢症状が誘導されるという新しい実験モデルが開発された。その発症メカニズムが、CD4^+Th2型細胞のホーミングおよびサイトカイン産生能から解明されつつある。
2)粘膜免疫機構の破綻に基づく炎症性ならびにアレルギー性腸管傷害の発生機序。
(1)粘膜免疫機構による粘膜バリアーの破綻に基づいた炎症性、アレルギー性消化管障害の発症機序とその修復過程において腸管局所に存在する好中球マクロファージ、樹状細胞がその局所で過剰かつ持続的な免疫反応を誘導していることを免疫組織学的に明らかとした。
(2)炎症性腸管障害および消化管アレルギーの病因病態では、VIP等ニュー口ペプチドならびに視床下部・下垂体・副腎軸を介した神経内分泌系による制御が破綻していることを明らかにした。
(3)好中球がこれらの病変の初期の粘膜上皮細胞傷害とアポトーシスを誘導し、その除去により活動性病変を改善することを明らかにした。
