血中酸化LDL定量法による新しい体外診断薬への展開

Research Abstract	われわれは、酸化LDLをより定量的に評価することを目的として、DLH3抗体を利用した酸化LDLの高感度定量法を開発した。この方法で、硫酸銅処理した酸化LDLを標準物質とすると、酸化LDLの蛋白量で10mg程度まで直線的に測定できる。多数の健常者、およびいくつかの疾患患者についての測定結果をまとめてみると、健常者では、LDL5μgあたり0-52ngの硫酸銅処理酸化LDLと同等の反応性が得られた。一方、人工透析患者では約8倍、家族性LCAT欠損症では、健常者の約3.5倍と有意に高い値が得られた。しかしながら、血漿総コレステロール値が有意に高い高脂血症患者ではLDL酸化値の上昇は認められない。さらに、約100名の循環器疾患患者、人工透析患者の血漿LDL酸化値を測定したところ、これら疾患患者の約半数以上は高値を示し、なかには著しく高い値を示す場合があった。すなわち、酸化LDL値はコレステロールとは独立した循環器疾患の危険因子である可能性が考えられる。
