ケモカインならびにレセプターの分析法の開発とその病態検査学的意義の検討

Research Abstract

本年度の主な研究成果は以下の通りである。
1)IL-4遺伝子導入細胞株の腫瘍拒絶過程において、所属リンパ節で内因性に産生されるインターフェロンγのみならず、ケモカインの一つである単球走化因子(MCP-1)が必須の役割を果たしていることを明らかにした。さらに、肺癌転移モデルにおいて、MCP-1がナチュラル・キラー細胞の活性を高めることによって、肺への転移を抑制することを明らかにした。
2)肝癌細胞株では、異なる2種類の転写因子であるPEA3とAP-1とが協調的に活性化されることによって、ケモカインの一つであるインターロイキン8(IL-8)遺伝子の転写が恒常的に起き、その結果恒常的に産生されることを明らかにした。さらに、C型肝炎患者血清中の種々のケモカイン濃度を測定したところ、血清IL-8濃度のみが、慢性肝炎・肝硬変患者に比べて、肝癌患者において選択的に上昇していることも確認した。これらのことは、IL-8が肝細胞の悪性形質転換時に肝癌細胞株から産生される可能性を強く示唆している。
3)サイトメガロウイルスによるIL-8遺伝子転写の誘導は、サイトメガロウイルスのintermediate early gene product 1によることを明らかにした。
4)糖尿病性腎症において、間質のマクロファージがMCP-1陽性に染まり、尿中MCP-1濃度も上昇していることを見出した。重症腎炎患者の腎臓における、ケモカインであるmacrophage inflammatory protein-1α(MIP-1a)・RANTESに対するレセプターである、CCR1とCCR5の局在を検討したところ、CCR1腸性細胞が間質病変に、CCR5陽性細胞がMIP-1αを介して糸球体病変、RANTESを介して間質病変の成立にそれぞれ関与していることを示唆する結果を得た。
