| Research Abstract |
私達は非ウイルス遺伝子治療法のベクターとして、合成ペプチドdiC_<18>-Gly-apoEを聞允した。LDL-受容体と結合するアポ蛋白Eの129-169のペプチド合成物のN末端をパルミチック酸でアシル化し、さらにグリシンでアルキル化し、よりコンデンスさせLDL受容体に結合しやすくした。この新ベクターにスカベンジャー受容体SR-Bl遺伝子アンチセンスを結合させ、その発現を抑制できるかを観察した。今回私達は、diC_<18>-Gly-apoEペプチドによるgene delivering systemを用いることにより、より臓器特異的にLDL受容体を介し、スカベンジャー受容体SR-BIアンチセンスを培養細胞核内に有効に導入できた。第一段階のデータとしてFITC標識スカベンジャー受容体SR-BIを効果的に実験細胞の核内に導入したことを認めた。現在同様の方法を用い、培養肝細胞、肝組織とのincubation実験、実験動物生体内での肝灌流実験に移行し、アンチセンス療法の特異性を生体内でも維持できるかを確かめている。
