Research Abstract |
We attempted to demonstrate the nuclear translocation of transcription factor, NF-x B in cultured human umblical vein endotherial cells (HUVEC) and in human monocytes reacted with the extracellular products from Streptococcus mitis and the heat-killed whole cells. Cytokines released from the ECP-or the whole cells-stimulated HUVEC and monocytes were also assessed. The results obtained were as follows. 1) The ECP exhibited strong cytotoxicactivity to HUVEC, and did not induce the nuclear translocation of NF-x B in HUVEC. The ECP could not also induce the release of TNF-α, IL-1β and IL-6 from HUVEC. 2) The ECP did not activate NF-x B in monocytes/macrophages, but induced the release of TNF-α, IL-1β and IL-6 from monocytes/macrophages. 3) The heat-killed whole cells induced the nuclear translocation of NF-x B, the expression of mRNA of cytokines as TNF-α, IL-1β and IL-6 in monocytes/macrophages, and the release of these cytokines.
