γδT細胞の発達分化と生体内生理的機能の解明

Research Abstract	新たに同定したマウス腸管リンパ組織であるクリプトパッチには胸腺外で発達分化して腸管粘膜最前線、すなわち腸管上皮層に分布する上皮細胞間T細胞(intestinalintraepithelial T lymphocytes:IEL)の前駆細胞が集積することを明らかにした(Science28:275-278,1998)。マウスIELにはαβ型T細胞抗原受容体(TCR)を保持するαβ-IEL(40〜70%)とγδ型TCR保持するγδ-IEL(30〜60%)が存在し、その大多数はCD8ααホモダイマーを発現する。さらに両IELは最終的な機能T細胞にまで分化しており、その細胞質内には多くの顆粒が存在するとともに、ex vivoでキラーT細胞(CTL)活性が検出される。 αβ-IELとγδ-IELの腸管粘膜局所、すなわちクリプトパッチでの発達分化機構の詳細は未解明であるが、本研究で得られた新知見によってαβT細胞が存在しないβ^<-1->マウスではγδ-IELのCTL活性やin vivoでのIFNγ産生が消失すること、これに対しβ^<dim>T細胞が少数存在するα^<-1->マウスではγδ-IELのCTL活性やin vivoでのIFNγ産生が逆に著しく亢進することすることが明らかとなった。一方、γδT細胞が存在しないδ-1-マウスのαβ-IELのCTL活性やin vivoでのIFNγ産生は正常であった。これらの結果は腸管粘膜でのγδ-IEL機能亢進がβ^<dim>T細胞に依存することを示しており、α-1-マウスに見られる炎症性腸疾患の機序解明に重要な新知見と考えられる(Proc.Natl.Acad.Sci.USA;in press)。
