Research Abstract	1) われわれは心不全患者、腎不全の血中glial-cell-line-derived neurotrophic factor(GDNF)が著明に増加していることをあきらかにした。GDNFは心不全・腎不全の成因や心・腎肥大・成長になんらかの重要な役割を果たしている可能性が示唆され、GDNFの動態や心・腎内局在を検討中である。 2) Hypoxia inducible factor(HIF)はvascular endothelial growth factor (VEGF)という血管新生増殖因子を増加し、心保護的に働くことが明らかになっている。Wistar-Kyoto ratでの心筋虚血再灌流時の心筋でのHIF発現レベルに対する長期間の運動(treadmill exercise)の効果を検討した。運動(-)虚血(-)群に比較して運動(-)虚血(+)群では心筋HIFmRNA発現は有意に増加していた。運動(+)虚血(+)群では、運動(-)虚血(+)と心筋HIFmRNA発現レベルに差異は認めず、本モデルにおいての虚血に対する長期的運動の効果はあきらかとはいえなかった。 3) われわれは5/6腎摘高血圧自然発症ラット(SHR)に運動(treadmill exercise)を長期的に行わせた際に尿蛋白減少作用、腎組織保護作用が認められることをあきらかにした。また、本モデルにおいて運動の腎機能保護作用およびアンジオテンシン変換酵素阻害薬enalaprilやアンジオテンシンII受容体拮抗薬losartanと運動の相乗作用が確認された。一方、エンドセリン受容体(ET-A)拮抗薬(FR139317)の血圧低下、腎保護作用は、本モデルにおいては認められず、長期的運動による作用を凌駕するものではないことが示唆された。
