Research Abstract |
1. We assessed the renal benefits of cicletanine (CIC), a furopyridine derivative drug with diuretic and antihypertensive properties, in diabetic spontaneously hypertensive Izmo rats (SHR) and Wistar-Kyoto rats (WKY) with renal impairment. CIC has a renal-protective action, which is not related to improvement of diabetes and improvement of high blood pressure in diabetic WKY and diabetic SHR. The actions might be due to the reduction of intraglomerular capillary pressure or protection of the renal glomerular vascular endothelial cell / mesangial cell injury through stimulation of PGI2 generation or elimination of free radicals, although the mechanism remains to be further investigated. 2. We assessed the renal effects of mild to severe chronic treadmill exercise in the spontaneously hypertensive rats (SHR) remnant kidney model of chronic renal failure (CRF). Chronic treadmill exercise significantly attenuated the increase in proteinuria and in serum total cholesterol levels intensity-dependently. Moderate-Ex significantly and effectively protected against increases in the percentage of focal glomerular sclerosis (FGS). Enalapril, losartan and amlodipine significantly decreased blood pressure, further decreased proteinuria and the percentage of FGS similarly in the rats with moderate Ex. These results indicate that exercise does not worsen renal function and suggest that exercise have renal-protective actions in this model of rats. Moreover the antihypertensive therapy has synergic renal-protective effects in this model of rats. 4.
