血管新生抑制因子(AIP)の遺伝子の解析と切除各種癌組織における発現の検討

Research Abstract	癌の進展に伴い,血管形成を抑制することは癌治療において非常に大きな意味があると考えられる.そこで,我々は,血管内皮細胞のHUVECの遊走能に着目し,その運動を制御する因子を見つけることにした.その手段として,機能性モノクローナル抗体を作成し,そのエピトープである遺伝子をクローニングしていくことにした.得られたHM7-3抗体は,実際にニワトリの受精卵を用いた実験で,血管新生をin vivoで抑制した.また,ラットの角膜を用いた実験でも角膜内への血管新生を高度に抑制した.この抗体のepitopeは分子量135K,105Kの糖蛋白質と考えられ,遺伝子クローニングを行ったところ,目的遺伝子の一部と考えられる511個の塩基配列からなるcDNAをクローニングした.この構造はGenbankに登録されていなかったため,未知物質と考えられ,我々はこれをAngiogenesis inhibiting protein(AIP)と名付けた.一時,このwhole cDNAと考えられる2571個からなるcDNAをクローニングできた. しかしながら,これをトランスフェクションしてみても,うまく血管新生制御を行うことが不可能であった.このため,むしろこの一部の蛋白構造に修飾された糖鎖構造がこのような生理的現象をもっている可能性が示唆された.現在,この抗体を用いて抗体アフィニティカラムを作製し,AIPの蛋白質部分と,糖鎖部分と精製してその構造を決定中である.
