| Research Abstract |
1. これまでにRACE法によりCloningしたPTEN2(仮名)のcDNA fragmentをBAC libralryを用いてscreeningした。新規の遺伝子と思われたPTEN2はChromosome9上のpeseudogeneであった。そこで、本研究課題の方針をPTEN2を中心とした解析から、PTENおよびその下流で働くAKT-BAD、ならびにIntegrin-linked kinaseの発現解析に変更した。
2. 現在、食道がん、胃がん、乳がんのcell lineおよびprimary carcinomaでPTENのmutation screeningを行っている。乳がんおよび胃がんの12%でmutationを見つけた。
3. PTENのmutationのあったcell lineで、real-time semiquantative PCRを用いてAKTのmRNAのoverexpressionを確認した。Laser capture microdissection systemにてprimary carcinomaのがん細胞をselectiveに採取し、AKTのmRNAの発現量を解析したところ、scirrhous carcinomaの浸潤部で発現の更新をみた。PTENのmutationのある例で特に強く発現しており、BADの発現量には変化がなかった。PTENのチロシンリン酸化機構は、scirrhous typeのcarcinoma cellのsurvivalに関連し、この種のがんでのapoptosisの回避に重要な役割を担っている可能性が考えられた。
4. 現在Intergrin-linked kinaseとPTENならびにAKTおよびBADの発現についてCell lineとmicrodissectionを行った材料で解析を検討している。
