| Research Abstract |
種々の解酒生薬のスクリーニング試験の結果,月桂樹(クスノキ科植物Laurus nobilis,葉)の抽出エキスに顕著なアルコール吸収抑制活性を見出した.
アルコール吸収抑制活性を指標に探索した結果,costunolideやdehydrocostus lactoneなどのα-methyl-γ-lactone環を有するセスキテルペン類が活性本体として単離された.種々の誘導体や関連セスキテルペンなどの活性を比較検討して活性発現の必須構造を解明するとともに,胃排出能抑制作用に関与した作用機序を見出した.
また,アルコール吸収抑制活性が認められた月桂樹セスキテルペン類は,合成医薬品よりも強力なアルコール性胃粘膜損傷に対する保護作用を示すほか,肝保護作用やNO産生抑制活性が認められた.さらに,これらの活性とセスキテルペン構造に関しても興味深い相関のあることが判明した.
このほか,解酒生薬の山人参(セリ科植物Angelica furcijuga,根茎),野菊花(キク科植物Chrysanthemum indicum,花),沢瀉(オモダカ科植物Alisma orientale,根茎)および茶(ツバキ科植物Camellia sinensis,種子)にアルコール性胃粘膜損傷の抑制作用,肝保護作用,NO産生抑制作用のあることを見出し,新規活性成分として,クマリンhyuganin類(山人参),ジアリルヘプタイドbetulaplatoside類(白樺),セスキテルペンkikkanol類(野菊花),トリテルペンalismaketone類(沢瀉),およびサポニンassamsaponin類(茶)を単離し,それらの化学構造を明らかにするなど平成11年度計画をほぼ達成した.
