Research Abstract |
The aim of this research is to identify genes regulating fetal mouse germ cell development including fate. determination of primordial germ cells (PGCs), regulation of growth and survival of PGCs, regulation of the initiation of meiosis and regulation of differentiation into gonocytes. For this purpose, I attempted to isolate genes showing developmental stage-specific or sex-specific expression in fetal germ cells or their supporting somatic cells by using subtraction cDNA screening. To isolated genes involved in determination of PGCs, I performed subtraction screening with purified migrating PGCs and blastocyst containing pluripotential cells, and succeeded to identify genes expressed in PGCs but not in pluripotential cells and somatic cells. I also succeeded to identify geiies specifically expressed in male but not in female gonadal somatic cells by using 12.5 dpc male and female genital ridges. Those genes are possibly involved in regulation of sex specific differentiation of fetal germ cells including initiation of meiosis.
