炎症細胞浸潤を媒介する接着分子P-セレクチンの新規機能ドメインの解析

Research Abstract

活性化血管内皮細胞および活性化血小板に発現するP-セレクチンの特異的なリガンドとして、白血球上に発現するムチン様糖タンパク質P-selectin glycoprotein ligand-1(PSGL-1)が知られている。最近の報告によると、PSGL-1のN末端近傍がシアリルルイスX様糖鎖修飾およびチロシン残基の硫酸化修飾を受けており、この2つの修飾が同時に存在することで、P-セレクチンとの結合に高い親和性をもたらしていると考えられている。われわれは、抗ラットP-セレクチンモノクローナル抗体を作製し、PSGL-1との結合に関与するP-セレクチン上の機能的部位の解析を試みた。その結果、P-セレクチンとPSGL-1との結合は阻害するがシアリルルイスXとの結合は阻害しないユニークなモノクローナル抗体C215を樹立し、この抗体がP-セレクチン上の8アミノ酸よりなるWADNEPNNという配列を認識することを明らかにした。さらに、この8アミノ酸よりなる合成ペプチドがP-セレクチンとPSGL-1の結合を特異的に阻害すること、およびシアリルルイスX糖鎖非依存的にPSGL-1遺伝子導入細胞と結合することを明らかにした。これまで、シアリルルイスX糖鎖結合ドメインのみがP-セレクチン上の機能ドメインと考えられてきた。しかし、以上の結果より、P-セレクチン上のアミノ酸76から83番のWADNEPNNがPSGL-1の認識に関与する新規機能エピトープである可能性が示唆された。
