老化モデルとしてのミトコンドリアDNAノックアウトマウスの樹立と応用

Research Abstract	(1)mtDNAノックアウトマウスの樹立にほぼ成功し,論文を投稿中である。この論文に関しては,最近査読者から極めて好意的なコメントをもらい,現在それに基づき修正中である。またこのマウスの作製と病態モデルとして利用に関し,特許の準備中である。特許の出願に関しては,すでに筑波大学の発明委員会の許可が得られ,筑波リエゾン研究所を通して出願手続中である。 (2)生きている細胞内のミトコンドリア間に相互作用がありmtDNAとその産物がミトコンドリア間で交換できることを発見した。このことにより,酸化的ストレスの激しいミトコンドリア内に存在するmtDNAに突然変異が蓄積しても,直ちにエネルギ-欠損にならないことが明らかになった。 (3)実際にアルツハイマ-病で亡くなった老化したヒトの脳の剖検検体からミトコンドリアをmtDNA欠損細胞に導入してその機能を調べた所,エネルギ-産生に関して何も問題のないことが明らかになり,この現象は上記の相互作用の存在により説明できることを明らかにした。これまで,運動に伴ってミトコンドリア内に大量に生じる酸化的ストレスにより,mtDNAの破壊や突然変異が誘導されることから,激しい運動を続けることは健康にとって好ましくないという仮説が提唱され,この仮説は現在も議論の対象となっている。今回の結果は,仮に変異が蓄積されても,少なくともmtDNAの相互作用により,核DNAの場合の論理で推察されるような好ましくない結果は生じないことが明らかになった。
