| Research Abstract |
交付申請書に記した実験実施計画に基づき以下の実験を行った。
1. 当大学動物実験施設.においてSPF環境下に飼育された、インスリン依存性糖尿病(IDDM)を自然発症するNODマウスについて、その単離膵島及び浸潤細胞を用いてFas、Fas-L、perforinの発場をRT-PCR法により検討した。現在得ている予備的な知見では比較的早期よりFas、perforinの発現を検出している。NODマウスでは、浸潤リンパ球を含まない膵島の単離は困難であり、現状ではこれらの発現がいずれの細胞に由来するものかは明確ではない。今後さらなる検討を行う予定である。
2. Fas-FasL系に対する中和抗体によるIDDM発症阻止効果についての実験を行った。現在までの成績では、中和抗体によって糖尿病の発症は阻止されていない。今後は抗体投与下の膵の組織学的分析を行う予定である。さらに抗体投与を受けたマウスについて各種サイトカインの発現の検討を行い、Fas-FasL系が免疫寛容の破綻、膵B細胞障害の現場およびTh1/Th2バランスの調整といかに関わるか免疫学的検討を行う予定である。
3. Fas抗原欠損NOD 1pr/1prマウスより単離した膵島を糖尿病未発症および発症後のNODマウスに.移植する実験については現在これを行っており、今後得られた知見をもとに糖尿病の進展性と可逆性について検討解析する予定である。
