Research Abstract	超伝導マグネット、リニアモーターカー、磁気共鳴画像化診断装置(MRI)等の新しい電磁気素材・機器の登場により、現代人は嘗て経験したことのない高磁場とくに低周波交流高磁場に遭遇する機会が増えつつある。が、高磁場が生体に一体どのような影響を与えるかについては未だ不明な点が多い。そこで本研究では、低周波交流高磁場の生体作用とくに遺伝子機能への影響を解明することを目的とする。本年度研究は予定通り順調に進行し、以下の成果を得た。 1. ポリ(dA)鋳型、オリゴ(dT)プライマー、[^3H]TTPまたは[^<35>S]dCTPを含むヌクレオチド基質とKlenow酵素からなる試験管内DNA合成反応を60ヘルツピーク値1テスラの高磁場暴露・非暴露下に行った。その結果、(a)DNA合成速度、(b)DNA合成の忠実度とも有意な差がなく、これはKlenow酵素が3′→5′校正エキソヌクレアーゼ活性を欠いた場合にも同様であった。(c)1テスラ変動高磁場は、がんや神経疾患の発症と関連づけられているDNAの滑り合成にもほとんど影響をおよぼさなかった。 2. レポーター遺伝子(lacZ)にミスマッチ変異を有するファージDNAをrepair-proficientなHeLa細胞抽出物と1テスラ高磁場の存在・非存在下でインキュベート後、repair-deficientな大腸菌変異下部(mutS)に移入してX galを含む培地にプレーティングし、形成される青色/無色プラークと混合プラークの比率からミスマッチ修復効率を算定した結果、高磁場印加の有無で差がなかった。したがって、ピーク値1テスラまでの高磁場は変異原性に乏しいと推定された。
