| Research Abstract |
糖鎖は細胞の表面の大部分を覆っており、細胞間や細胞一基質問相互作用に重要な役割を果たしていると考えられている。しかし、糖蛋白質糖鎖の機能解明はあまり進んでいない。その、大きな理由の一つとして、細胞表面や組織中で発現している糖蛋白質糖鎖の系統だった解析方法がないことが挙げられる。われわれは糖蛋白質糖鎖の系統だった解析法をほぼ自動化することに成功したので、今回ヒト脳の分析を行った。10%ホルマリン固定ヒト病理標本正常部位(遺伝的に独立した4例、40-45歳、男女各2例で、各々前頭葉中前頭回、前頭葉白質、被角、アンモン角、小脳皮質)20検体をアセトン沈殿し解析に用いた。蛍光標識後にノイラミニダーゼ処理し、シアル酸を除去した後に糖鎖構造解析のための2次元HPLCを行った。その結果脳の各部位においてN-結合型糖鎖発現パターンに個人差はほとんどないことが明らかとなった。ピークの高さに違いのあることはあっても、発現しているN-結合型糖鎖の種類は全く同じであり、非常によく一致していることが分かった。しかしながら同一個体でも領域が異なれば発現パターンも少し異なっていた。今後、年齢変化について検討する。
