Research Abstract	COE形成基礎研究費「類人猿の進化と人類の成立」では、主として京都大学霊長類研究所の類人猿研究グループを1)行動、生態、社会、2)形態、古生物学、3)脳科学、4)遺伝学、分子生物学の四班にまとめ、研究計画の初期にはそれぞれの班の研究を深め、後半にそれらの連携を計ることを考えている。まず生態班では、アフリカの類人猿、チンパンジー及びゴリラの種内亜種間、種間の生態、社会、行動上の変異を比較・検討した。本年度はとくにチンパンジーの政治行動、狩猟行動、ゴリラのコミュニティの構造について分析した。亜種間の遺伝的差異や父子判定の解析を計画しているが、チンパンジー等を一時的にせよ捕獲し採血することは不可能である。食物のシガミカス、糞、尿、体毛等の試料を、ウガンダのカリンズ、ギニアのボッソウの森で採集した。またチンパンジーの亜種の分布域境界に近いカメルーンでネストから体毛を集めた。他方、インドネシア、カリマンタン島でテナガザルの行動観察を行い、同時に糞、体毛を採集した。現在遺伝的変異性の非常に高いマイクロサテライトの分析を開始した。彼らの血縁関係の解明は世界に先駆ける研究となる。形態班ではチンパンジーとニホンザルについて、pQCT装置を用いて骨密度の年齢変化を調査した。これら二種で生涯最高骨密度はそれそれ15〜20才,約10才であること等、骨リモデリング研究のための基礎的知見を得た。類人猿研究を目指して、導入設備の二波長X線骨密度測定装置を用い、ニホンザルで予備的研究を行った。また、ガボン国にて、西ローランドゴリラの形態・遺伝資料を収集した。他方ミャンマー国において同国と共同で化石の調査を行い、真猿類のおおもとの種と考えられる化石を発見した。またタイ国でも調査し、1500-2000万年前の地層から哺乳類、脊椎動物の化石を発見した。この時代はアジアに類人猿が分布し始めた時代に相当し、将来類人猿化石の発見が期待される。中国に関しては北京の古脊椎動物・古人類研究所で、雲南省出土の類人猿化石標本を調査した。脳科学では類人猿の脳で侵襲的研究はできないため、類人猿の脳のモデルとしてマカカ属サルの脳内から複数のニューロン活動を記録し、購入した多ニューロン分離システムによって単一ニューロン活動に分離したうえで、ニューロン相互の情報連絡様式を解析した。また核磁気共鳴映像装置、MRIにより麻酔下のチンパンジーの脳のイメージング分析を行い、脳の左右差を検討している。今後研究所で飼育している他のチンパンジーについて行い、個体差を調べる予定である。その際は言語訓練の有無等に注目したい。他方マカカ属サルを用い陽電子放射断層撮影、PET法により分析した。視覚刺激により視覚野の興奮が記録された。本年度購入のマルチラベルカウンターを使用し、霊長類の神経成長因子(NGF)と脳由来神経栄養因子(BDNF)の酵素免疫測定法の開発に成功した。現在各種霊長類の血清や脳組織におけるこれらの栄養因子の定量を行っている。またこれらの栄養因子の受容体の脳内分布とその発達を調べた。チンパンジーの妊娠末期の流産胎児を用い、大脳皮質における神経細胞の分布とBDNF含有細胞について調べた。遺伝班では類人猿からの非侵襲的な試料として尿からのDNAがPCR法の試料として有効であることを明らかにし、実際試料採集も行った。植物食から肉食に至る類人猿の食性の多様化の要因を分子レベルで明らかにするため、各種類人猿で胃の消化酵素ペプシノゲンの成分分析と遺伝子の構造解析を進めた。今年度購入した核酸抽出システムの使用によりプラスミドの抽出が自動化されたことにより、テナガザル、オランウータンについてcDNAクローニングが終了した。ミトコンドリアDNA(mtDNA)のCOの偽遺伝子と考えられるDNA断片を発見し、系統分布を検索した結果、類人猿、真猿類で存在し、原猿類と霊長類以外の系統群では存在しないことを確認した。購入したシーケンサーを用い約600塩基の配列決定を行った結果、mtDNACOとの相同性は70%程度と推定された。現在FISH法で核遺伝子化したサイトの同定を進めている。新たに開発したこの分子標識を利用し、分子レベルと染色体レベルの双方から類人猿を中心とした霊長類系統群の比較研究を計画している。
