Research Abstract	1.神経変性疾患発症と神経機能に関わる分子の同定 (1)GDNF-RET系の神経細胞の生存、分化に関わるシグナル伝達系の解析 GDNF-RET系により活性化される細胞内シグナル伝達系を解析した結果、RAS/MAPK系、PI3-K/AKT系、JNK系、p38MAPK系すべてRETのチロシン1062を介して活性化され、PLCγはチロシン1015を介して活性化されることを明らかにした。さらにGDNFにより発現誘導される複数の新規遺伝子をディファレンシャルディスプレイ法にて同定することに成功した。 (2)筋萎縮性側索硬化症(ALS)の分子病態分子インデックス法を用いてALSの脊髄前角において発現変化の見られる遺伝子についてクローニングを行った。発現の変化みられた遺伝子のうち2つの新規遺伝子の全長cDNAの塩基配列を決定した。このうち1つはRing-finger/IBRmotifを有する蛋白をコードしていた。 (3)ミッドカイン受容体の同定とシグナル伝達 13日マウス胎仔の膜画分からアフィニティーカラムを用いてMK結合蛋白を得た。プロテインシーケンサーで解析した結果、low density lipoprotein receptor-related protein(LRP)がMK結合蛋白として同定された。 (4)糖転移酵素遺伝子の神経機能における役割 GM2/GD2合成酵素遺伝子ノックアウト、GD3合成酵素遺伝子ノックアウトマウスの解析の解析により、複合型ガングリオシドの適切な発現が神経系組織の維持と再生、免疫系、生殖系などの機能遂行に必須であることが示された。 2.癌の増殖、浸潤、転移の分子機序とその制御 (1)v-Srcキナーゼによる癌の浸潤・転移の分子機序について v-Srcおよびv-Crk癌化細胞において、Ras-MEK1-MAPKシグナル伝達系がMMPの分泌と活性化に決定的なシグナルである事を同定した。SHPS1と呼ばれる糖蛋白質遺伝子の発現は多くの癌において特異的に抑制されている事を発見した。更に、ヒト乳癌で高率に発現が亢進しているシグナル伝達因子遺伝子MAWDを同定し、その強発現はMAPKの活性化を介して細胞を癌化することを発見した。 (2)活性型RETを介したシグナル伝達の解析多発性内分泌腫瘍症(MEN)2B型、家族性甲状腺癌(FMTC)ではRETの変異がキナーゼドメインに検出されている。MEN2B型の変異2種とFMTC型変異3種をRETに導入し解析した結果、トランスフォーミング活性など両者の生物学的活性に違いのあることを明らかにした。 (3)悪性腫瘍とMK ヒトのがんが全般的にMKを強く発現し、その進展に伴ってMKの発現が増大していくことがいくつかのがんで明らかになった。早期がんの検出にもMK値の測定が利用できる可能性が示唆された。 3.神経変性疾患、悪性腫瘍のモデル動物の作製 (1)MEN2A型変異RET遺伝子導入トランスジェニックマウスの作製 MEN2A型変異RET遺伝子をモロニー白血病ウィルスLTRに結合し、トランスジェニックマウスの作製を行い、甲状腺髄様癌を必発する系を確立した。 (2)球脊髄性筋萎縮症(SBMA)モデル動物の作成 CAGリピート病のモデルマウスとしてアンドロゲンレセプター遺伝子(tAR)を組み込んだトランスジェニックマウスを作成した。ヒトのSBMAに認められるのと同様の核内封入体が神経系細胞に認め、運動能力も有意に低下するマウスが確立できた。 4.遺伝子の効率的な細胞への導入法の確立 (1)シャペロン遺伝子導入による神経変性疾患治療法の開発 Neuro2a細胞にtruncated AR(CAG)97-GFP vector(tAR97-GFP)を各種シャペロン(Hsp70,Hsp40,Hsdj)と共に遺伝子導入すると、Hsp70/Hsp40群及びHsp70群で強い核内凝集体形成抑制・apoptosis抑制効果を認め、トリプレットリピート病の治療法開発に応用できる可能性を示した。 (2)MKアンチセンスRNAおよびMKプロモーターを用いた癌細胞の増殖抑制マウスMKmRNAの二次構造を予測し、塩基対を形成しない部位に対してのアンチセンスRNAが、マウス大腸がん細胞CMT-93のMK分泌をほぼ完全に抑え、同時にCMT-93細胞の増殖能と、軟寒天中でのコロニー形成も阻害した。
