| Research Abstract |
小児気管支喘息は高頻度にアトピーと関連している。喘息小児は種々のアレルゲンに対するIgeが亢進しているが、イエダニやカビが最も多いものである。筆者らはイエダニに感作している喘息同胞とその両親47家系、197人に対して、全ゲノムを対象とした連鎖解析をResearch Genetics社のHuman Screening Setを用いて全染色体上に平均10cM間隔に存在する398遺伝マーカーについて遺伝子型を決定した。多点連鎖解析はMAPMAKER/SIBを用いた。最大ロッド値4.8を染色体5q31-q33のインターロイキン12B付近に得た。その他に最大ロッド値4q35(MLS=2.7),6p22-p21.3(MLS=2.1),12q21-q23(MLS=1.9),13q14.1-14.3(MLS=2.0)が連鎖が示唆された領域であった。本研究で得られた連鎖が示唆された領域の多くは、白人においても連鎖が示唆されている。この研究ではインターロイキン12B遺伝子を含む領域が喘息に関わっているということと、喘息に関わっている遺伝子は人種を越えて共通であることが示唆された。
精神分裂病に関しては、日本人の精神分裂病患者を発端者とする37家系、121人、57同胞対を対象に、全染色体上に平均10cM間隔に存在する遺伝マーカーについて遺伝子型を決定した。連鎖は精神分裂病の易罹病性を反映している探索眼球運動に対して、QTL解析法を用いて行った。同胞間での形質の差に基づく解析により、連鎖の可能性のある領域として、第2染色体の2q12-q14領域(p=0.006)、第5染色体の5q13-q23領域(p=0.001)、第11染色体の11q13-q14領域(p=0.008)、第12染色体の12q15-q21領域(p=0.009)、第16染色体の16q22-q23領域(p=0.002)、第17染色体の17q24-q25領域(p=0.0002)、第22染色体の22q11領域(p=0.002)が得られた。
