Research Abstract

出芽酵母Tom1蛋白はC末にHect-ドメインを持つユビキチンライゲースである。欠失株は温度感受性を示し、高温にシフトすると、核分裂の開始、核構造維持、mRNAの核外輸送など、多面的な欠損が観察される。tom1の温度感受性を抑圧する変異株を単離したところ、Hsp70をコードするpdr13,Hsp70スーパーファミリーをコードするsse1や、DnaJ-ファミリーをコードするzuo1変異株であった。sse1とzuo1変異との間では遺伝学的関連は見い出されなかったが、pdr13とzuo1変異株は低温感受性増殖を示し、general stress responseが構成的に発現していた。そこで、Pdr13とZuo1タンパクが、Hsp70-DnaJパートナーであるかを検証した。2者は2-ハイブリド法による結合が確認された。さらに、それぞれにタグを付加し、免疫沈降実験を行ったところ、共沈した。従って、Pdr13とZuo1は、新たなHsp70-DnaJパートナーであり、遺伝子発現に関与していると示唆された。
