Research Abstract	本研究は、シグナル伝達における蛋白質複合体の詳細な構造・機能解析を目指して、以下の2点を目指している。(1)抗ニワロリリゾチーム抗体HyHEL10Fv-エリスロポエチン受容体キメラ蛋白質を受容体のリガンド認識機構研究のモデルケースと捉え、HyHEL10Fv-受容体キメラ蛋白質と抗原リゾチームとの複合体のX線結晶解析を行う。(2)異種抗原認識能を有するHyHEL10Fv変異体を用いて細胞増殖能の変化を調べ、さらに変異HyHEL10Fv-受容体キメラ蛋白質とリガンド複合体の構造解析を行い、分子認識能と細胞表面からの情報伝達の機構に迫る。今年度の実績は以下の通りである。(1)前年度構築したヒト並びにマウス由来のエリスロポエチン受容体について、その細胞外ドメイン遺伝子をクローニングし、大腸菌を用いた高発現系並びに抗体分子に準じた、不溶性顆粒からの高効率な巻き戻し系を用いて、結晶構造解析を初めとする各種物理化学的解析に十分対応できる試料を調製し、マウス由来受容体については得られた試料を用いた結晶化条件の検討をより詳細に行った。また、変異体を作製し、等温滴定型熱量測定、表面プラスモン共鳴を用いて、高親和性ペプチド並びにエリスロポエチンを用いて相互作用を詳細に解析した。(2)前年度構造解析を行ったFv変異体と受容体のキメラ蛋白質について、前年度構築した発現系を用いて、卷き戻し系を構築し、結晶構造解析を初めとする各種物理化学的解析に十分対応できる試料を調製し、各種物理化学的解析を行う。(3)既に確立してあるFvとニワトリリソチームとの複合体の結晶化条件をもとに、抗体-受容体キメラ蛋白質の構造機能解析において用いる変異抗体について、抗原との複合体の結晶を引き続き作製し、高エネルギー加速器研究機構の放射光装置で精密なデータ収集を行い、相互作用について詳細な考察を行った。
