| Research Abstract |
1.Paracoccus denitrificansにおけるPHA合成系遺伝子の発現様式の解析
n-ペンタノールまたはメタノールを唯一炭素源にして、N源豊富培地からN源枯渇培地へ菌体を培地交換し、RNAレベルおよびタンパク質レベルでのPHA合成・分解に関連する遺伝子群(合計5種類)の発現様式を検討した。それぞれの遺伝子に対して興味深い結果が得られた。
2.菌体内PHAデポリメラーゼの解析
Paracoccus denitrificansの菌体内に存在する3HB oligomer hydrolaseおよびPHB depolymeraseの両者に関して、精製と諸性質を検討した。前者については、その生合成は構成的であった。また、精製酵素の諸性質はZoogloea ramigeraのそれと比較的類似していた。後者については、既知の遺伝子(phaZ)産物とは別の酵素の存在を、菌体の粗酵素溶液の硫安分画、陰イオン交換カラムクロマトグラフィーの溶出プロフィールから認めた。この新規酵素の生成物は3HB monomerであった。このような、3HB monomerを生成するPHB depolymeraseを分離したのは世界で初めてである。
