| Research Abstract |
これまでに85種類以上の精巣生殖細胞特異的遺伝子cDNAをクローニングして、解析を行ってきた。平成11年度はGsg3(actin capping protein alpha),Haspin,Tektin,Sperizinについて報告した。平成12年度はさらに以下の遺伝子について解析を行った。
精巣特異的succinyl CoA:3-oxo acid CoA transferase(Scot-t):ミトコンドリアに局在し・エネルギー代謝の鍵となる酵素。体細胞型のScot-Sがヒトや豚で報告されているが、その精巣型アイソフォーム。遺伝子はイントロンレスで、体細胞型Scot-S遺伝子からレトロポジションにより生じたと考えられる。発現は精巣半数体精子細胞特異的で、体細胞型にはない酵素活性の特異性を持つものと思われる。
Phosphatidylcholine transfer protein(PCTP)-like(PCTP-L):精巣での発現は半数体精子細胞特異的で、精子のエネルギー代謝や先体反応に必須であるphosphatidylcholineの細胞内輸送に重要と考えられる。精巣特異的ornithine decarboxylase antizyme(OAZ-t):細胞増殖や分化に必須であるポリアミン濃度を制御する。体細胞型のOAZ-Sに対する精巣型アイソフォーム。半数体精子細胞で体細胞型の消失とともに入れ違いに発現する。
haploid-specific alanine-rich acidic protein located on chromosome-X(halap-X):塩基性アミノ酸が豊富な核蛋白質で、精巣半数体精子細胞特異的に発現する。クロマチン凝縮に関与するとみられる。
また、30種類の遺伝子についてはマウス染色体マッピングを行い、ほとんどすべての染色体上に散在すること、これらのマウス遺伝子にはほとんどヒトオーソローグが存在することを明らかにした。
