| Research Abstract |
アポトーシス時には、細胞内コンパートメントを越えてシグナル伝達が行なわれるが、我々は、特にミトコンドリア、細胞質、核の間でのシグナル伝達機構の解明に取り組んできた。種々のアポトーシスシグナルは最終的にミトコンドリアに集約され、カスペース活性化因子シトクロムcなどの細胞質への漏出を惹起し、カスペースが活性化される。カスペースは種々の細胞質内たんぱくを切断し、アポトーシスに特徴的な構造変化を誘導する。たとえば我々の同定したAcinusはカスペースによる切断の後、クロマチン凝縮を誘導する。
本年度の成果は以下の通りである。
Acinusによるクロマチン凝縮機構およびその正常時での機能を解明する目的でAcinusノックアウトマウスの解析を継続し、ホモマウスが示す胚致死性は発生の遅延によること、またこの現象が、構成する胚細胞が示す低い増殖率に起因する可能牲を示唆した。また、Acinus結合たんぱくを解析し、RNAプロセッシングに関わる因子、RNPS1などに結合することを示した。
