| Research Abstract |
我々は,平成10,11年度で,アンドロゲン受容体のAF-1蛋白質を用いたGST-pulldown実験でアンドロゲン受容体共役因子異常症患者者で特異的に欠損する90kDaの蛋白質を同定した。
平成13年度はこの90kDaタンパク質をクローニングする目的で,アンドロゲン受容体AF-1をbaitとして,酵母Two-Hybridシステムを用いてスクリーニングした結果,941アミノ酸からなる既知のタンパク質を得た。このタンパク質はその分子内にTPR(Tetratrico Peptide Repeat)モチーフを19回反復するという特徴を有するため仮にTPRタンパク質と名付けている。このタンパク質は,レポーターアッセイでアンドロゲン受容体の転写活性化能を3〜5倍増加させ,GST pulldown assayでアンドロゲン受容体AF-1領域に直接結合することが判明しているが,エストロゲン受容体の転写活性化能は増加させない。
このTPRが本疾患の病因であるかどうかを明らかにするため現在このcDNAの12のprimerを作成し,PCRでmutationを検討中である。
更にDNAチップを使い正常細胞に比べ,本疾患患者で欠損している遺伝子を同定中である。
