| Research Abstract |
ラット褐色細胞腫由来株PC12細胞をもちいて、アポトーシスシグナル蛋白分子とPLC、PLD活性に対する局所麻酔薬の作用を検討した。
テトラカインはMAPKファミリーのリン酸化を介してアポトーシスを誘発し、ERKの活性化はアポトーシスから細胞を保護し、JNKは細胞死を誘発するように働き、Na^+チャネルやL-型Ca^<2+>チャネルはこのシグナル伝達に関与していないという結果を得た。
BradykininによるPLD活性化はtetracaine前処理により濃度及び時間依存的に抑制された。しかし、Na^+チャネルブロッカーのTTX、L-type Ca^<2+>チャネルの阻害薬のnifedipine前処理ではbradykininによるPLD活性化に影響しなかった。Ca^<2+>フリーの細胞外液で処理した細胞ではPLDの活性化は観察されなかった。0.5mM tetracaine前処理では、チロシンリン酸化に影響しなかった。また、ERK経路の阻害するPD98059は、bradykininによるPLDの活性化を部分的に抑制した。PLD活性化はprocaine>lidocaine>bupivacaie>tetracaineの順で抑制され、tetracaineがGTP結合蛋白質刺激剤であるGTPγSによるPLD活性化を濃度依存性に抑制した。以上の結果から、局所麻酔薬が好中球細胞において、PLD活性化因子の膜転移を阻害して、その活性化を抑制することを示した。
