| Research Abstract |
タバコ培養細胞BY-2を分裂期に同調化し、ゴルジ小胞を単離を試みた。ゴルジ小胞であるという判断基準は1)電子顕微鏡観察による大きさが約0.1umである、2)キシログルカンを含む、とした。その結果、比較的高純度の膜分画を得ることができた。ゴルジ小胞は細胞板形成時には微小管によって赤道面に輸送されると考えられているので、単離したゴルジ小胞と微小管の相互作用を調べた。加えたゴルジ小胞の約30%が微小管に結合した。また、モータータンパク質活性を調べるためにゴルジ小胞をガラス上に固定し、微小管を添加してin vitro motility assayを行った。しかし、微小管の運動は見られなかった。これらの否定的結果は、ゴルジ小胞の純度が低い、単離の際にゴルジ小胞に結合した微小管付随タンパク質あるいはモータータンパク質が解離した、などが原因であると考えられ、今後単離方法の改良を進めなければならない。一方、融合機構を調べるためにゴルジ小胞を分裂後期に同調化したBY-2細胞miniprotoplast extractに加えた。その結果、アニリンブルー染色される直径2-3um塊状の構造が形成された。しかし、形成の確率が低いことや細胞板に比較すると小さいことから、今後、細胞周期や微小管の有無などを検討してよりよい試験管内再構成系を作らなければならない。
