| Research Abstract |
今年度は主としてデキストラン加水分解酵素について研究を行い,下記のような結果を得た。デキストランのα-1,6結合を認識し,エンド型にこれを水解する3種の酵素(glucodextranase,isomalto-dextranase,isomaltotrio-dextranase)のうち,isomaltotrio-dextranaseの遺伝子を解析し,一次構造を明らかにするとともに,大腸菌での発現にも成功し報告した。glucodextranaseについても同様な研究を行い,一次構造を明らかにした。この酵素は既に我々が明らかにしたエンド型デキストラナーゼのisomalt-dextranaseおよび上記のisomaltotrio-dextranaseとは相同性がなかったが,驚くべき事に,Clostridium由来のglucoamylaseと37%の相同性を示した。さらにこの傾向は,glucoamylaseの活性部位と推定される領域と顕著であった。α-1,6結合水解酵素glucodextranaseとα-1,4結合水解酵素glucoamylaseが類似の構造を持つことから,今後両者のキメラ酵素を作成して解析することにより,基質認識の重要な機構が明らかになると期待される。
