リポ蛋白リパーゼの動脈硬化発生における役割-トランスジェニックウサギを用いた研究

Research Abstract	平成11年度はヒトリポ蛋白リパーゼを過剰発現するトランスジェニックウサギfounderlineの作製並びにその解析を行った。現在までに全身に発現する(β-actin prmoterを使用)3羽のトランスジェニックウサギの解析をしたところ、多臓器でヒトLPLmRNAを発現しており、ヘパリン注射後のLPL血中濃度は(L01)395、(L04)157、(L17)654ng/dl(ヒト健常者130ng/dl)で、正常ウサギより3倍に近く高い活性が認められた。また、L17トランスジェニックウサギを用いて血漿リポ蛋白プロファイルおよび脂質を測定した。トランスジェニックウサギの血漿総コレステロール、中性脂肪、HDL-C値はコントロールに比べ、いずれの場合も減少しており、血漿のVLDLも著しく低下していた。L17トランスジェニックウサギの系統についてすでに確立しており、繁殖も行っている。現在動脈硬化並びに脂質代謝の研究に必要な多数のF1ウサギ作製を目指して交配を開始したところである。また、神戸大学塩見雅志先生の協力でLPLトランスジェニックウサギとLDL受容体欠損ウサギとの交配も開始した。その他に、マクロファージに特異的に発現する(マクロファージスカベンジャー受容体promoterを使用)リポ蛋白リパーゼを過剰発現するトランスジェニックウサギの作製も試み、1羽のfounderウサギが成功した。このウサギの解析も現在進行中である。
