| Research Abstract |
1.突然変異のない野生型のWT1遺伝子は,白血病のみならず,肺癌などの固形がんでも高発現しており,WT1アンチセンスオリゴマー処理により,細胞増殖が抑制された。
2.WT1アンチセンスオリゴマー処理により,白血病細胞K526は,cell cycleのG_2/Mでarrestした。
3.WT1遺伝子には,4種類のtranscript(17AA+1KTS+,17AA+/KTS-,17AA-/KTS+,17AA-/KTS-)が存在する。この4種類のtranscriptを別々に定量できる測定系を確立した。この測定系を用いて,白血病細胞でのWT1 transcriptを測定したところ,17AA+/KTS+がmajorであった。
4.myeloid progenitor cell32DにWT1遺伝子を導入し,その発現をON/OFFできる系を作った。この系を用いて,WT1遺伝子の発現をONにすると,アポトーシスが抑制された。
5.WT1タンパクと結合する新規結合タンパクをクローニングし,WTIPと名付けた。WTIPは,splicesomeに局在し,splicing factorと考えられる。
