Research Abstract	転写因子HNF-1αはインスリン合成や分泌に関する重要な遺伝子を転写調節している。本年度では、HNFカスケードに属するHNF-1α標的遺伝子の収集を重点的に行った。この遺伝子プールから有力候補を選択し、次年度以降のMODY患者の遺伝子解析に供する予定である。 (1)膵β細胞株(MIN6)発現遺伝子のEST化とHNF-1α過剰発現による誘導遺伝子の獲得マウスMIN6細胞からmRNAを抽出しcDNAライブラリーを作成した。In vivo excision 法によりライブラリー全体をプラスミドに変換した後、無作為に5,000個以上のクローンを選択してEST化した。得られたEST配列についてデーターベースサーチを行ない、重複しないESTパネルを作成中である。次年度中には一万個を終了してマイクロアレイ化する。 MIN6細胞でHNF-1αを効率よく発現させるためにアデノウイルス発現系を用いた。アデノコンスラクトを同細胞に感染させてHNF-1αを過剰発現させると、標的遺伝子mRNA全体のレベルを上昇させることができる。本年度は、このアデノ発現ベクターの構築に成功した。 (2)対応するヒト膵ラ氏島ESTパネルの作成ヒト膵ラ氏島cDNAライブラリーから既に1万個のESTを獲得している。上記と同様にデータベースサーチを行ない、重複しないヒトESTパネルを作成した。このパネルは、HNF-1α過剰発現で発現レベルが変化するマウス遺伝子に対応するヒト遺伝子の獲得に用いる。
