Research Abstract |
In order to clarify the mechanism of the posttranslational activation of RubisCO in prokaryotes, we used a thermophilic hydrogen bacterium, Hydrogenophilus thermoluteolus. And the following results were obtained. (1) Gene product of cbbQ and/or cbbO.which locates downstream of the structural genes for RubisCO, is involved in the RubisCO activation. (2) Through analysis of CD spectra, activation of RubisCO is accompanied by strautural change of the protein. (3) CBBQ catalyzes ATP hydrolysis. The reaction required Mg^<2+> and was inhibited by ADP.The characteristics are similar to those known for RubisCO activase. In contrast that RubisCO activase hydrolyzes only ATP, CBBQ hydrolyzed not only ATP but also UTP, GTP, CTP, and TTP. (4) Analysis of gene cluter for Calvin-Benson cycle was completed. The gene arrangement was clarified as follows ; cbbLSQOYA-pyk-cbbRFPTGK.
