| Research Abstract |
小児の自己免疫性腸炎(AIE)の自己抗原をコードするcDNAをbaitとして酵母two-hybrid法でplacenta cDNA libraryからMCC(mutated in colon cancer)癌抑制遺伝子のホモローグMCC-2をクローニングした。さらに、腎臓および大腸のcDNAライブラリーからWnt pathwayのkeyとなる複数の蛋白がクローニングされた(未公表)。そこで、AIE-75にこれらの蛋白が結合すると酵母のADE2レポーター遺伝子が転写される系を作製した。この系ではAIE-75蛋白に変異があればADE2レポーター遺伝子が転写されないために酵母は赤くなる。このように酵母の色彩変化を指標としてcDNAの変異を検出する系を作製している。現在、これらのアッセイ系はフレームシフト変異とナンセンス変異の検出は正しく行えるが、ミスセンス変異についてはまだ不十分で改善の余地がある。AIE-75結合蛋白の多くはシグナル伝達経路のメンバーであるので酵母内にこの経路を再構築してこの経路が動くか否かによって全ての変異を検出できる系を作製中である。
