| Research Abstract |
腸管壁在神経と平滑筋に間の介在するカハールの介在細胞(カハール細胞)は消化管の自立的蠕動運動のペーシング機能を担っている。私共は、c-kit遺伝子に機能喪失性突然変異を持つミュータントラットではカハール細胞の減少によってペーシングが低下して消化管蠕動運動が低下すること、また、実際の臨床において慢性突発性偽腸閉塞症患者で腸管筋層のカハール細胞が減少していることを明らかにし、消化管運動異常症における蠕動運動の異常にカハール細胞のペーシングの異常が関与することを明らかにした。消化管運動異常症を治療するためには、従来の腸管神経を標的とした治療薬ではなく、カハール細胞は、腸管平滑筋細胞と共通の前駆細胞から分化すると考えられているが、c-kit以外にカハール細胞に特異的に発現する分子は未だ明らかになっていない。私共は、カハール細胞に特異的に発現する分子を明らかにし、そのペーシング機能調節機構を明らかにするため、温度感受性突然変異SV40大型T抗原遺伝子を導入したマウスの小腸からカハール細胞の培養を確立した。マウスのカハール細胞と腸管平滑筋細胞のcDNAライブラリーを用いてcDNA差分化法によりcDNAライブラリーから平滑筋細胞由来のmRNAと相補的なインサートを有するクローンをハイブリダイゼーションおよびアビジン/ビオチン・トリックにより除去し、得られたカハール細胞に特異的に発現するクローンについてシークエンスを行い塩基配列を決定し遺伝子の同定をおこなう予定である。
